ABSTRACT:
Collagen, the primary structural protein in the skin’s dermal extracellular matrix (ECM), plays a critical role in supporting firmness, hydration, and elasticity. Its synthesis and organization are regulated by dermal fibroblasts, whose activity is influenced by extracellular matrix interactions and environmental stressors. Accelerated collagen degradation largely mediated by matrix metalloproteinases (MMPs) and oxidative stress contributes to skin aging. Bioactive phytocompounds with antioxidant and anti-inflammatory properties have emerged as enhancing collagen synthesis while inhibiting extracellular matrix (ECM) degrading enzymes. The Timeless Radiance Anti-Aging Cream (Anvaya Ayurveda, Greenspace, India) previously demonstrated cytocompatibility and modulatory effects on Aquaporin-3 in human dermal fibroblasts (HDFs). The present study designed to evaluate its effect on collagen gene expression using an in-vitro Human dermal fibroblasts (HDF). Cytotoxicity was evaluated by MTT assay, confirming non-toxicity up to 1000µg/mL. Two non-cytotoxic concentrations (500 and 250µg/mL) were subsequently assessed for gene expression using RT-PCR with GAPDH as an endogenous control. Treatment with Timeless Radiance resulted in a significant, dose-dependent upregulation of collagen gene expression (1.38±0.06-fold), comparable to the enhancement observed with the hyaluronic acid standard (2.02±0.01-fold). The multi-component botanical blend rich in antioxidants, anti-inflammatory agents, and hydrating bioactives provides a broader spectrum of biological benefits indicate that the Timeless Radiance Anti-Aging Cream supports collagen biosynthesis and may contribute to dermal rejuvenation through molecular modulation of ECM-related pathways.
